Health Secretary Matt Hancock has announced that Hastings and Rother will be moving into Tier 3 from 12:01am on Saturday.
The rest of Sussex will remain in Tier 2.
Mr Hancock has also confirmed the highest band of restriction will come into force in Bedfordshire, Buckinghamshire, Berkshire, Peterborough, the whole of Hertfordshire and Surrey (with the exception of Waverly).
Also going into Tier 3 will be Portsmouth, Gosport and Havant in Hampshire.
Bristol and North Somerset will be moving down from Tier 3 into Tier 2 on Saturday.
Herefordshire is going from Tier 2 to Tier 1.
The Health Secretary said:
"People must be vigilant and keep this virus under control.
"We've come so far, we mustn't blow it now.
"Across the world, cases are rising once more.
"In Europe, restrictions are being reintroduced. No one wants to do that but where those are necessary they must be put in place to protect the NHS and safe lives.
"Cases are up 46% in the last week in south east of England, it is necessary to apply Tier 3 much wider area of the east and south east."
The restrictions will come into force before the relaxation over Christmas from Wednesday (December 23).
Three households will be allowed to mix for five days, regardless of tiers.
